Essentially it's all started by a plague which wipes out all men (bar one- Yorick) on earth. With only women remaining and just about half of the population wiped out, planes fall out of the sky, infrastructure falls apart and gangs start to form (Daughters of The Amazon, in particular- who go around destroying sperm banks). [Small Spoilers here] They try to put things back together but without men to create babies it won't last long, so they attempt some cloning. As well as that, the last remaining man (immune to the plague) runs about trying to go from the USA to Australia to find his girlfriend and make babies.
So, first you'd have to develop the plague and successfully wipe out all men, then destroy sperm banks and then cloning facilities, and finally find and kill Yorick before he reaches Australia. (Keep in mind, he'd only leave once he was the last man alive) You could possibly even throw in attempts from some of the remaining ladies to bed Yorick that you have to prevent. If you're successful there, good times, but if the cloning succeeds, you would then need to enhance your original plague to kill the clones.
